Glacios2 200kV Cryo-TEM

The Glacios™ 2 is a 200 kV, CFEG Cryo-Transmission Electron Microscope (Cryo-TEM) that can be configured for Single Particle Analysis (SPA) high-resolution data acquisition, sample quality assessment, Tomography, MicroED, and other applications.

Features & Specifications

  • E-CFEG: The low-energy-spread cold field emission gun facilitate an energy spread as low as 0.3 eV to deliver high-contrast and atomic resolutions beyond 0.2 nm.
  • Ceta-D Camera 200 kV: It is the latest Thermo Scientific CMOS camera used in experiments such as MicroED and imaging.
  • Selectris with Falcon 4i for 200 kV: Falcon 4i is an advanced direct electron detection camera with an increased internal frame rate of 320 fps. Selectris is an energy filters designed for optical stability and reproducibility during on extended automated data collection.
  • Optics and apertures: Automatic Aperture System, with:
    • C2 condenser apertures 20, 50, 70, 150 μm
    • Objective apertures 30, 70, 100 μm
    • With Phase Plate option selected: Phase Plate, 70, 100 μm
    • Selected area apertures 10, 40, 200, 800 μm
    • With MicroED option selected: 10, 40, 100, 200 μm
    • C-TWIN Lens
  • Smart EPU with Quality Monitor: EPU is an automated data acquisition software by Thermo Fisher. It is also used in sample screening and optimization and to setup multi-grid data acquisition sessions.
  • MicroED Package: Although the microscope is primarily used for SPA workflow in the beginning, the MicroED Package contains all components required to automatically collect diffraction data on small, dose-sensitive crystals.
  • TEM Tomography Data Acquisition Software: The software is used to set tilt series data acquisition.
  • Tomo Live: Used to perform quality monitoring of tilt series data and real time reconstruction into 3D volumes.

Vitrobot Mark IV System

Used to plunge freeze the liquid samples onto the cryo-EM grids. The liquid samples may contain proteins, nucleic acids and other biomolecules suspended in suitable buffer or water. After the plunge freezing in liquid ethane, the grids are clipped and transferred to the microscope for screening, optimization and data acquisition.

Features & Specifications

  • Peltier controlled heating/cooling
  • Ultrasonic controlled humidification
  • Maintains ambient humidity to 100% relative humidity
  • Two-sided blotting with adjustable blot force
  • Adjustable wait time and drain time options

PELCO easiGlow™ Glow Discharge Cleaning System

The glow discharge system is used to activate the TEM grids and make it more hydrophilic using air.

Features & Specifications

  • Adjustable settings to control plasma current (0 to 30 mA), vacuum pressure (1.1 to 0.20 mbar)
  • Adjustable glow discharge and hold time
